Biorepository

The main CALERIE protocol calls for samples of blood, urine, and tissue to be procured for repository storage in the following quantities:

Sample type

Visits at baseline, 12 months
and 24 months

Visits at 3 months, 6 months, and 18
months

Serum  10 ml  4 ml
Serum (OGTT) 4 ml
Plasma (EDTA)

9 ml

4 ml
Plasma (citrate) 1 ml

RNA

Extracted from whole blood at BL, 12 months and 24 months
DNA Extracted from whole blood at BL, 12 months and 24 months
Urine (no additive) 18 ml
Urine (with boric acid) 18 ml
Skeletal muscle ~ 45 individuals Note: a small subset of CALERIE individuals had all three samples
Adipose tissue ~ 45 individuals Note: a small subset of CALERIE individuals had all three samples

Some of the aliquoting of samples was be performed at the field centers; the remainder occurred upon arrival at our laboratory.  Regardless of location, the personnel entrusted with this responsibility were appropriately trained and demonstrated competence in the application of the aliquotting procedure.  Most of the aliquots for storage measured either 0.5 ml or 1.0 ml volumes.

CALERIE SAMPLES: Numbers of participants with at least one stored sample of the type and at the times indicated.
